The string “PYTHON” has six characters,
numbered 0 to 5, as shown below:

So if you wanted “Y”, you could just type
"PYTHON"[1] (always start counting from 0!)
fifth_letter = Y

print fifth_letter

I think that, if you study the example in the comments and probably the instructions as well, you can get a better idea of what the exercise wants you to do. (Seems like you kind of ignored the instructions completely and should therefore look closer at them rather than be asking)
