<em> and <strong> or <i> and <b>?

Is there a difference between and ? And between and ?

There is a difference.

<i> and <b> are used to change the appearance of text (regularly done through CSS now), where <em> and <strong> have semantic meaning while also changing the appearance of text.

Take for example, this heading:

Our Product is Fifty Times More Reliable!

<h2>Our Product is <i>Fifty Times</i> More Reliable!</h2>

vs

Our Product is Fifty Times More Reliable!

<h2>Our Product is <em>Fifty Times</em> More Reliable!</h2>

Visually, they both look the same. However, the second has semantic meaning in that the “Fifty Times” is emphasized. Screen readers may also read it differently; emphasizing while reading the second heading but not the first.
